Vaccines are among the most effective methods to guard against infectious diseases, but their efficacy varies from person to person. As people age, their immune systems tend to weaken, leading to a reduced response to vaccinations. Researchers now believe that slowing this immune decline could enhance vaccine effectiveness.

Scientists at NDORMS investigated whether spermidine, a naturally occurring compound found within human cells and certain foods, might boost the immune response in older adults. Their research was published in Aging Cell.

During the COVID-19 pandemic, older adults represented the majority of fatalities. While vaccines substantially decreased severe illness, many seniors produced fewer antibodies and had weaker T-cell responses compared to younger individuals. Understanding why this disparity occurs has become a key focus for researchers.

The study involved 40 healthy volunteers over 65 who had already received three doses of a COVID-19 vaccine. Participants were randomly assigned to take either a daily spermidine supplement or a placebo for 13 weeks. Both the participants and the researchers were blind to the group assignments until the trial concluded.

Overall, the supplement was safe and well-tolerated. Notably, scientists identified a subgroup of individuals who responded poorly to the vaccines despite receiving three doses. Their immune cells exhibited signs of biological aging, such as DNA damage and cellular senescence, indicating the cells had largely lost normal function.

In these low responders, spermidine supplementation correlated with increased antibody production, healthier B-cell activity, and a better ability to neutralize different COVID-19 variants. The researchers also observed that spermidine appeared to stimulate autophagy—a natural cellular process that recycles damaged components and promotes cell health.

These results imply that poor vaccine responses in some cases could be linked to accelerated aging within immune cells rather than just chronological age. If future research confirms this hypothesis, healthcare providers might eventually develop therapies aimed at delaying immune aging, improving vaccine outcomes.

However, many questions remain. The small size of this trial means the findings are preliminary, and larger studies are needed before any routine clinical recommendations can be made. It’s also important to explore whether spermidine has similar effects with other vaccines, like the flu shot.

The study’s strengths include its randomized, placebo-controlled design and its focus on uncovering the biological mechanisms behind inadequate vaccine responses. Its main limitation is the limited number of participants, underscoring the need for more extensive research.

Until then, these findings should be viewed as promising but preliminary. For those interested in health, emerging evidence suggests that vitamin D may help reduce inflammation, and vitamin K could potentially cut the risk of heart disease by about one-third. Additional recent studies highlight new ways to combat excessive inflammation and identify foods that may trigger it.
